Experimental Studies on T-cell Activation of the Herba Lobeliae Chinensis

Abstract
Objectives This study was carried out for the purpose of knowing the effect from T-cell activation of the C57BL/6 mouse by the extraction from a extract of herba lobeliae chinensis(EHLC.). Methods We examined the influence of EHLC on T cell proliferation by using T cell gained in spleen and thymus gland of C57BW6 mouse. Also we examined cytotoxicity through SBR assay and immunocell through FACScan
Results :
The results were obtained as follows:
1. In order to know the effect of the cytotoxicity from extraction of EHLC, we had to examine the safe density of that on lung fibroblast cells of the mouse(mLFCs). EHLC did not show cytotoxicity against mLFCs.
2. In the proliferous effect of thymus T cells, water extracts of EHLC played an important role on the proliferating thymus T cells compared with control group at low density.
3. EHLC promoted the CW(B7-1) and Thl cytokines especially IL-2 gene expression, but decreased CTLA-4, IFN-V, CD28 and Th2 cytokines IL-4 gene expression.
4. EHLC up-regulates CD44¢¥CD69¢¥CD25¢¥, CD4¢¥CD25¢¥, CD8¢¥CD25¢¥, CD3¢¥CD80¢¥ expression but not CD3^(+)CD28^(+) by anti-CD3-stimulated T cells.
Conclusions : above results indicates that EHLC play an important role in the activati on of autoreactive T cells by promoting Thl cytokine(I1-2) and dec reasing CTLA-4, IFN-Y, CD28 and Th2 cytokines II-4 gene expressi on, by up-regulating cell surface molecule(CD80, CD25, CD44). The refore further deep studies shoud be accomplished about its effects.
